Its principle is based on the polymerization of 7-methylguanosine 5 ' diphosphate ( m ~ 7GDP ) on an oligonucleotide primer in the presence of polynucleotide phosphorylase ( PNPase ), followed by selective elimination of m ~ 7G under mild conditions .
